ALCAM, activated leukocyte cell adhesion molecule, influences the aggressive nature of breast cancer cells, a potential connection to bone metastasis.

چکیده
INTRODUCTION ALCAM, activated leukocyte cell adhesion molecule, is connected to the progression of certain solid tumours and has been shown to be a prominent feature for tumours that subsequently developed bone metastasis. The present study investigated the biological influence of ALCAM on breast cancer cells in connection with bone biological environment. MATERIALS AND METHODS Suitable breast cancer cells were transfected with either the ALCAM expression construct or anti-ALCAM transgene, to create sublines that had differential expression of ALCAM. The growth, migration and invasion of the cells were evaluated in the presence or absence of matrix proteins prepared from human bones. RESULTS ZR-751(DeltaALCAM) (ALCAM knockdown) and MDA-MB-231(ALCAMexp) (overexpressing ALCAM) were constructed. MDA MB-231(ALCAMexp) cells showed a slower rate of growth compared with control cells. However, in the presence of bone matrix proteins, MDA MB-231(ALCAMexp) showed a significantly reduced rate of growth, p<0.01 vs. control cells. In contrast, ZR-751(DeltaALCAM) cells grew faster compared with control cells. MDA MB-231(ALCAMexp) displayed a significantly reduced (p=0.012) and ZR-751(DeltaALCAM) cells significantly increased invasiveness (p=0.02) vs. their respective controls cells. In an ECIS-based cell migration assay, MDA-MB-231(ALCAMexp) cells showed marked reduction in migration. Inclusion of bone matrix proteins therefore further reduced the migration speed of MDA MB-231(ALCAMexp) cells. CONCLUSION Loss of ALCAM in breast cancer cells facilitates the invasive behaviour of breast cancer and high levels of ALCAM in the cells have a suppressive role in the aggressive nature of breast cancer cells.

Expression Levels of Activated Leukocyte Cell Adhesion Molecule (ALCAM/CD166) in Primary Breast Carcinoma and Distant Breast Cancer Metastases
INTRODUCTION Activated Leukocyte Cell Adhesion Molecule (ALCAM/CD166) gained increasing attention regarding tumorprogression and metastatic spread in breast cancer. The aim of this study was to examine ALCAM expression levels in primary breast cancer and distant metastases of the same patient within 29 autopsy cases to better understand the underlying mechanisms of metastases and the role of ad...
متن کاملExpression of the cell to cell adhesion molecule, ALCAM, in breast cancer patients and the potential link with skeletal metastasis.
The activated leukocyte cell adhesion molecule (ALCAM) is involved in cell migration and adhesion. Decreased levels of ALCAM expression in breast cancer tissue are known to correlate with poor prognosis. The current study specifically investigated the ALCAM expression in tumours which developed skeletal metastasis. Fresh frozen primary breast cancer tissues (n=234) and non-neoplastic mammary ti...
متن کاملActivated Leukocyte Cell Adhesion Molecule (ALCAM) in Saudi Breast Cancer Patients as Prognostic and Predictive Indicator
BACKGROUND Activated leukocyte cell adhesion molecules (ALCAMs) play an essential role in tumor metastasis and are higher in some patients with breast cancer. AIM This study aimed to evaluate ALCAM as an early diagnostic biomarker for breast cancer and how it compares with other markers. SUBJECTS AND METHODS One-hundred and sixty-one women were selected for this study. They were divided int...
متن کاملMolecular and Cellular Pathobiology ALCAM/CD166 Is a TGF-b–Responsive Marker and Functional Regulator of Prostate Cancer Metastasis to Bone
The dissemination of prostate cancer to bone is a common, incurable aspect of advanced disease. Prevention and treatment of this terminal phase of prostate cancer requires improved molecular understanding of the process as well as markers indicative of molecular progression. Through biochemical analyses and loss-of-function in vivo studies, we demonstrate that the cell adhesion molecule, activa...
متن کاملAttenuation of melanoma invasion by a secreted variant of activated leukocyte cell adhesion molecule.
Activated leukocyte cell adhesion molecule (ALCAM/CD166/MEMD), a marker of various cancers and mesenchymal stem cells, is involved in melanoma metastasis. We have exploited a secreted NH(2)-terminal fragment, sALCAM, to test the hypothesis that ALCAM coordinates tissue growth and cell migration. Overexpression of sALCAM in metastatic melanoma cells disturbed clustering of endogenous ALCAM and i...
